The Northern Irishman hit a four-under par 67 on day three to climb to the top of the standings, with a score of 13 under, at Congaree Golf Club.
Spaniard Jon Rahm is second with South Korean Kyoung-Hoon Lee and American Kurt Kitayama.
Victory would almost certainly see McIlroy regain the world number one spot for the first time since 2020.
The four-time major winner, who has held the No. 1 spot in the world for the past eight streaks, can also topple Scottie Scheffler at the top with a second-place finish if the Masters champion finishes outside the top 35.
Scheffler is currently tied for 47th at normal going into the final day.
American pair Aaron Wise and Taylor Moore are tied for fifth with less than 10 while England’s Tommy Fleetwood is tied for ninth just two more strokes behind.
